ABSTRACT

Diarrhea is characterized by increased bowel movements and loose and even watery stools. Its occurrence and development have been proved by modern research to be closely related to the imbalance of intestinal flora. Traditional Chinese medicine (TCM) pays a special attention to syndrome differentiation in treating diarrhea. Exploring the TCM syndromes of diarrhea is of great significance to the formulation of TCM diagnosis and treatment scheme of diarrhea and the improvement of clinical curative effects. There exist many similarities between TCM theory and micro-ecological theory concerning diarrhea. With the deepening of intestinal flora research, the significance of intestinal flora in TCM syndrome research has been increasingly highlighted. The close correlation of intestinal flora with the occurrence and development of diarrhea has provided new ideas of deducing syndrome and selecting prescription based on intestinal flora. This paper summarized the relationship between TCM syndromes of diarrhea and microscopic indexes such as immune response, neurotransmitters, brain-gut peptide, and proteins and analyzed the intestinal flora characteristics related to six common TCM syndromes of diarrhea. Meanwhile, based on the theory of deducing syndrome by prescription, namely deducing the syndrome by the efficacy of prescription or its medicinal components, the correlation between TCM syndromes of diarrhea and intestinal flora was indirectly verified, so as to identify the research direction of correlation between intestinal flora and TCM syndromes in the future. Numerous studies have shown that the TCM syndromes of diarrhea were highly correlated with the microscopic indexes such as inflammatory cytokines, neurotransmitters, and proteins. Syndromes and dysbacteriosis both resulted from pathogenic factors acting on the body, which were summarized from different angles. Different TCM syndromes corresponded to specific objective indicators of intestinal flora. Intestinal flora has the potential of being an internal material basis for powerfully revealing the TCM syndromes of diarrhea in the future.

ABSTRACT

BACKGROUND: Circulating RNA (circRNA) regulates various bioactivities in cells. A better understanding of the exosomal circRNA can provide novel insights into the pathogenesis and treatment of Graves' disease (GD). We aimed to profile the differentially expressed circRNAs (DEcRs) in plasma exosomes of patients with GD and speculate and probe the functions of the DEcR by comprehensive bioinformatics analyses. METHODS: Serum exosomes were isolated from five primary GD patients and five healthy controls via ultracentrifugation. After verification with transmission electron microscopy, exosome samples were subjected to microarray profiling using human circRNA microarrays. Two up-regulated and two down-regulated DEcRs were selected for validation in plasma exosomes from 20 GD and 20 healthy control participants using reverse transcriptase-quantitative polymerase chain reaction (RT-qPCR). The circRNA/microRNA/mRNA interaction network was then assembled and the analysis of the Gene Ontology and KEGG (Kyoto Encyclopedia of Genes and Genomes) pathways was utilized to predict the potential functions of the DEcR associated genes. RESULTS: There were 15 DEcRs revealed in primary GD cases. The intronic circRNA hsa_circRNA_000102 was confirmed as an up-regulated component in plasma exosomes from patients with GD. The circRNA/microRNA/mRNA interaction network unveiled the most potential targeting microRNAs of hsa_circRNA_000102 and its associated genes. The functional analyses predicted involvement of hsa_circRNA_000102 associated genes in pathways of immune system activation, such as viral infection and interferon-beta signaling. CONCLUSIONS: hsa_circRNA_000102 is a differentially up-regulated plasma exosomal circRNA in patients with GD. Our study highlights multiple pathways, particularly virus infection and interferon-beta signaling, for mediating immune activation in Graves' disease.

@#Objective To derive the general equation of the probability distribution of identity by state (IBS) score among biological full sibling pairs by calculating STR allele frequency. Methods Based on the Mendelian genetics law and the hypothesis that parents of biological full siblings(FS) were unrelated individuals, the IBS score and corresponding probability of different genotype combinations in the offspring when unrelated individuals of different genotype combinations give birth to two offsprings were derived. Results Given fi (i=1, 2, …, m) as the frequency of the ith allele of a STR locus, the probability of sharing 2 alleles(p2FS), 1 allele(p1FS) or 0 allele (p0FS) with biological full sibling pairs on the locus can be respectively expressed as follows: p2FS= 14 ×[1 + 2∑im= 1 fi2 + 2(∑im= 1 fi2)2 -∑im= 1 fi4] , p1FS= 12 ×[1 +∑im= 1 fi2 - 2(∑im= 1 fi2)2 - 2∑im= 1 fi3 + 2∑im= 1 fi4] and p0FS= 14 ×[1 - 4∑im= 1 fi2 + 2(∑im= 1 fi2)2 + 4∑im= 1 fi3 -3∑im= 1 fi4] . The sum of p2FS, p1FS and p0FS must be 1. As for the multiple genotyping system that contained n STR loci, IBS scores between biological full sibling pairs conform to binomial distribution:IBS~B(2n, π1). The population rate π1, can be given by the formula:π1= 1n∑ln= 1 p2FSl + 21n∑ln= 1 p1FSl . Conclusion The alternative hypothesis in biological full sibling testing is that two appraised individuals are biological full siblings. The probability of the corresponding alternative hypothesis of any STR locus combination or IBS score can be directly calculated by the equations presented in this study, and the calculation results are the basis for explanations of the evidence.

ABSTRACT

Objective To prepare the Elderly Functional Constipation(FC) Health Education Scale, and verify its reliability and validity. Methods The Elderly FC Health Education Scale was prepared by qualitative interviews, literature review, and Delphi method. The elderly patients who were over 60 years old and at Chinese PLA 181st Hospital were recruited by the purposive sampling method from January 2013 to August 2015.The diagnostic criteria of RomanⅢwas used to diagnose FC, the data of preliminary investigation and large sample test was used to form the formal scale, and its reliability and validity were further verified. Results The Elderly FC Health Education Scale was compiled with 6 dimensions and 28 items, and the Cronbach alpha coefficient which was the internal consistency reliability of the 6 factor was 0.965; the correlation coefficient analysis of equality reliability Kendall tau-b rank and various index variables score were positive correlation significantly. Both item level content validity index and scale level content validity index of the content validity were as a result of 1. The structure validity of the cumulated variance contribution ratio of the 6 factors were 60.15%. All factor loading coefficients between the items were more than 0.5, which indicated the fitting was good. Conclusions The reliability and validity of the Elderly FC Health Education Scale are good, and the scale may be used as a tool to prevent the elderly FC health education, and also be applied to the elderly FC patients in self-management and continue nursing after leaving hospital.

ABSTRACT

Objective To learn the toxic effects on rats repeatedly exposed to dichloromethane by dermal route at the same dose for 28 days.Methods Seventy two Sprague Dawley rats were divided into six groups:a solvent control group (oliver oil),four exposure groups (0.75,1.5,3.0 and 6.0 g/kg·BW)and a recovery group.Each group included six female and six male rats.The exposure groups were dermal exposed to DCM at 1.5,3.0 and 6.0 g/kg·BW,6 hours/day,5 days/week for four weeks.The recovery group was dermal exposed to 6.0 g/kg BW of DCMduring the same period then recovered for 2 weeks.Clinical signs including body weight and food consumption of each group were observed.After rats were sacrificed,the hematological and serum biochemical parameters were determined.Histopathological examination was performed on selected tissues for all animals.Results The male rats in test groups including four exposure groups and a recovery group,showed decreased body weight gain.Neutrophilia and lymphocytopenia were determined in females of 3.0 and 6.0 g/kg BW groups and males of 1.5,3.0 and 6.0 g/kg BW groups by blood routine test (P <0.05).The mean levels of serum ALT and AST significantly increased in males of 3.0 and 6.0 g/kg BW groups (P <0.05).Only the mean level of serum TG in males of 6.0 g/kg BW group decreased (P <0.05).No significant differences of these indexes were shown in female rats of all exposure groups compared to control group.The exposure -related significant increases on the incidences of epidermis and dermis lesion,hepatocellular degeneration and necrosis were observed in test groups compared to the control group.The rats of recovery group were exhibited normal percentages of lymphocytes and neutrophils,levels of ALT,AST and TG. The skin and hepatocellular lesion were improved to some extent, but not recovered exactly.Conclusion In a 4 -week repeated -dose toxic study,DCM induce dose -related effects in rats involving skin lesion and liver injury with adverse changes in body weight gain of male rats,part parameters of hematology and serum biochemistry .

ABSTRACT

This paper was aim to screen microorganisms with attenualed efficiency for Chinese medicine containing aristolochic acid A by liquid-state fermentation. Twelve Chinese medicine were detected by UPLC and aristolochic acid A was only founded in four species of Aristolochia, those were Caulis Aristolochiae Manshuriensis, Aristolochiae Radix, Aistolochia Contorta Bunge and Herba Aristolochiae Mollissima,but not in the others. With the four Chinese medicine containing aristolochic acid A as raw material, ten microorganisms were tested, and the content of aristolochic acid A was detected by UPLC. The results showed that one microorganism can decrease content of aristolochic acid A in all those four Chinese medicine.

ABSTRACT

Botrytis cinerea is a necrotrophic pathogen causing pre- and post-harvest diseases in at least 235 plant species. It manifests extraordinary genotype and phenotype variation. One of the causes of this variation is transposable elements. Two transposable elements have been discovered in this fungus, the retrotransposon (Boty), and the transposon (Flipper). In this work, two complete (Boty-II-76 and Boty-II-103) and two partial (Boty-II-95 and Boty-II-141) long terminal repeat (LTR) retrotransposons were identified by an in silico genomic sequence analysis. Boty-II-76 and Boty-II-103 contain 6439 bp nucleotides with a pair of LTRs at both ends, and an internal deduced pol gene encoding a polyprotein with reverse transcriptase and DDE integrase domains. They are flanked by 5 bp direct repeats (ACCAT, CTTTC). In Boty-II-141, two LTRs at both ends, and a partial internal pol gene encoding a protein with a DDE integrase domain were identified. In Boty-II-95, a right LTR and a partial internal pol gene encoding a protein with no conserved domains were identified. Boty-II uses a self-priming mechanism to initiate synthesis of reverse transcripts. The sequence of the presumed primer binding site for first-strand reverse transcription is 5’-TTGTACCAT-3’. The polypurine-rich sequence for plus-strand DNA synthesis is 5’-GCCTTGAGCGGGGGGTAC-3’. Fourteen Boty-II LTRs that contain 125-158 bp nucleotides and share 69.1 ~ 100 percent identities with the short inverted terminal repeats of 5 bp (TGTCA…TGACA) were discovered. Analysis of structural features and phylogeny revealed that Boty-II is a novel LTR retrotransposon. It could potentially be used as a novel molecular marker for the investigation of genetic variation in B. cinerea.

ABSTRACT

The plant hormone abscisic acid has huge economic potential and can be applied in agriculture and forestry for it is considered to be involved in plant resistance to stresses such as cold, heat, salinity, drought, pathogens and wounding. Now overproducing strains of Botrytis cinerea are used for biotechnological production of abscisic acid. An LTR retrotransposon, Boty-aba, and a solo LTR were identified by in silico genomic sequence analysis, and both were detected within the abscisic acid gene cluster in B. cinerea B05.10, but not in B. cinerea SAS56. Boty-aba contains a pair of LTRs and two internal genes. The LTRs and the first gene have features characteristic of Ty3/gypsy LTR retrotransposons. The second gene is a novel gene, named brtn, which encodes for a protein (named BRTN) without putative conserved domains. The impressive divergence in structure of the abscisic acid gene clusters putatively gives new clues to investigate the divergence in the abscisic acid production yields of different B. cinerea strains.

ABSTRACT

<p><b>OBJECTIVE</b>To establish the three-dimensional (3D) visible models of the anatomical structures of the anterior approach to the upper thoracic spine (UTS) for anatomic study and preoperative planning of the UTS.</p><p><b>METHODS</b>Sectional images from the superior margin of the first thoracic vertebral body to the inferior margin of the fifth thoracic vertebral body were acquired through the Chinese Visible Human Female (VCHF) database which was collected by the Third Military Medical University. These images were imported into Photoshop CS, cut automatically and converted into a JPEG format. Surface and volume reconstruction were performed by 3D Doctor 3.5 and Amira 4.0 software programs on an ordinary personal computer respectively.</p><p><b>RESULTS</b>The surface reconstruction model could be rotated at any angle and observed from any direction. And the reconstructed structures of the anterior approach to the UTS could be displayed individually or as a composite with any other selected structure. The volume reconstruction displayed abundant internal details of the reconstructed images in transverse, coronal, sagittal, and random oblique sections.</p><p><b>CONCLUSION</b>Three-dimensional visible models of the anatomical structures of the anterior approach to the UTS based on the sectional images of VCHF can clearly display the morphology, spatial orientation and adjacent relationship of every structure. These models are very helpful to the anatomy study and preoperative planning of this complex anatomic region.</p>

ABSTRACT

In the present research, patch-clamp whole-cell recording was used to study the developmental changes of the internal horizontal synaptic plasticity in layer II/III of rats' primary visual cortices. Pairing stimulation was used to induce long term potentiation (LTP) of neurons in layer II/III from layer II/III and layer IV. The data indicate that: (1) Responses of layer II/III neurons can be evoked independently at II/III-II/III and IV-II/III synapses by horizontal and vertical stimulations; (2) LTP can be induced from neurons in the layer II/III by horizontal tetanic stimulation at II/III-II/III synapses till postnatal day12 (P12, before eyes open); (3) Meanwhile, only short term potentiation (STP) at IV-II/III synapses can be induced by horizontal tetanic stimulation before eyes open; (4) After P12, a robust LTP at IV-II/III synapses can be induced by horizontal tetanic stimulation; (5) At P14, when vertical and horizontal tetanic stimulations were given to the same neuron, the LTP at IV-II/III synapses was weaker than that induced by vertical stimulation alone, suggesting that vertical synaptic modification was negatively regulated by horizontal inputs when two-direction synaptic inputs were presented at the same time; (6) Spontaneous responses of AMPA receptors (AMPARs) in the layer II/III neuron of rats' primary visual cortices are regulated by the development. The frequency of AMPARs-mediated postsynaptic currents was at a low level before eyes open, increased sharply at P12-P14, and slightly decreased after P18. And the amplitude of spontaneous AMPARs currents slowly decreased after P12. The results demonstrated that both the strength of horizontal synaptic modification and the effects of horizontal inputs on the vertical synaptic connection are regulated by the development. II/III-II/III synaptic communication has dual effects on the IV-II/III synapses, which may be involved in a competitive machinery of neural circuitry maturation and the formation of visual function columns.

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinical effect under highly active anti-retroviral therapy (HAART) in AIDS patients and for improving the curative effect and prognosis.</p><p><b>METHODS</b>Epidemiological method was used from five aspects to describe the post-treatment clinical symptoms of 181 AIDS patients in Suizhou, and to evaluate the change of virus load and immune function of 79 AIDS patients. Data was doubly recorded by Epi Data and database was set up by SPSS 13.0 for analysis.</p><p><b>RESULTS</b>The effective powers of anomal-fever, cough, diarrhoea, lymphadenectasis, weight drop, erythra, mycotic infection were 81.39%, 85.00%, 84.62%, 81.89%, 82.86%, 66.07% and 45.45% respectively. CD4+ T lymphocyte count rose obviously after treatment, with an averag of 276 x 10(6) cells/ml (65 x 10(6)-824 x 10(6) cells/ml), an 129 x 10(6) cells/ml increase in three months and was 294 x 10(6) cells/ml (102 x 10(6)-750 x 10(6) cells/ml) in six months. The count change of CD4+ T lymphocyte between 3 months and 6 months did not show sigificant difference. The number of deaths among drug withdrawals was 14, with a case fatality rate as 29.79%; while the number of deaths among non-drug withdrawals was 3, with the case fatality rate as 2.24%.</p><p><b>CONCLUSION</b>Results through this study showed that HAART could obviously improve the clinical symptom of AIDS patients, and to increase the number of virus load. Improving the compliance could also reduce the case fatality rate.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the anatomical structures, and depth and direction of needling at Jingming (BL 1), so as to provide anatomical basis for its clinical application.</p><p><b>METHODS</b>Forty-eight adult orbital specimens were observed by dissection.</p><p><b>RESULTS</b>When a needle was vertically inserted into Jingming (BL 1), the needle tip will past through the skin, subcutaneous tissue, medial palpebral ligament, medialis rectus and orbital adipose body. Above the body of the needle, there are ophthalmic artery, anterior ethmoidal artery and nasociliary nerve. The average distance between the skin at the punctured point and the anterior ethmoidal artery is (18.25 +/- 4.45) mm, with an angle of (12.5 +/- 5.5) degrees, and the average distance between the skin at the punctured point and the optic nerve tunnel frontal point is (43.37 +/- 7.84) mm.</p><p><b>CONCLUSION</b>To avoid bleeding caused by injuring the anterior ethmoidal artery, acupuncture at Jingming (BL 1) should avoid deeply inserting needled back-upwards and upwards, and the needling depth should not exceed 30.36 mm to avoid injury of the optic nerve tunnel frontal point.</p>

ABSTRACT

<p><b>AIM</b>To investigate the paclitaxel-loaded nanoparticles of poly(ethylene glycol)-b-poly(D,L-lactic acid) amphiphilic diblock copolymer (PMT).</p><p><b>METHODS</b>PMT was prepared by solid dispersion technique. The average size and size distribution were determined by dynamic light scattering (DLS). The morphology was characterized by transmission electron microscopy (TEM) and 1HNMR. The influences of the copolymer molecular weight and the paclitaxel-fed amount on PMT were studied. Therapeutic effect of PMT was studied on Kunming mice liver cancer H22.</p><p><b>RESULTS</b>PMT showed nanometer size and spherical morphology with core and shell. The sizes of PMT increased with increasing the molecular weight of the hydrophobic segment in PEDLLA or increasing the drug-loaded amount. The tumour inhibiting effect of PMT was similar with that of Taxol.</p><p><b>CONCLUSION</b>It will provide an experiment basis for the development of new kind of intravenous administration of paclitaxel.</p>

Aim To explore the effect of sesquiterpene lactone parthenolide(PL) on primary multiple myeloma(MM) cell and cell lines and its molecular mechanisms.Methods Fresh bone marrow cells from MM patients and MM cell lines KMM-1 and MM1S were used for in vitro culture study.The cell survival rate was examined by Trypan Blue exclusion.The change of nucleolus stained with DAPI was observed under fluorescent microscope.Reactive oxygen species(ROS) levels and mitochondrial membrane potential(MMP) were detected by flow cytometry.Results 2.5 ?mol?L-1 PL induced MM cell apoptosis and MMP was profoundly decreased in KMM-1 and MM1S cells.In contrast,normal lymphocytes cells were unaffected by the same conditions.KMM-1 and MM1S cells showed large increases of ROS levels 10 h after 2.5 ?mol?L-1 PL treatment.The free radical scavenger L-N-acetylcystein(L-NAC) pretreatment completely inhibited ROS generation,cell death and nuclear fragmentation by PL.Diphenylene iodonium chloride(DPI) pretreatment in a dose of 2.5 ?mol?L-1 strongly inhibited PL-mediated ROS generation,nuclear fragmentation in KMM-1 and MM1S cells.Conclusions PL strongly induces apoptosis in MM cell,while normal lymphocytes cells are completely unaffected by the same conditions.Since its proapoptotic activity appears to be mediated through induction of oxidative stress in MM cells,its possible mechanism is that NADPH oxidase activation is involved in PL-mediated ROS generation.
